In septic T cells, VDAC1 oligomers open a pore that leaks mtDNA and drives inflammation, and VBIT-12 shuts it down
Here's the kind of paper that keeps me hooked. A group at Shanghai Children's Hospital (Shanghai Jiao Tong University), led by Yiping Zhou, was digging into why sepsis makes immune cells go haywire. They hit mouse T cells with bacterial toxin and basically filmed the disaster in slow motion: the mitochondria got stressed, VDAC1 molecules clumped together on the outer membrane, and mitochondrial DNA started leaking out into the cell, where the cGAS–STING sensor mistook it for an invader and cranked up inflammation.
The kicker is the fix. When they blocked VDAC1 from clumping with VBIT-12, one of the oligomerization inhibitors I keep going on about, the mtDNA stopped leaking and the inflammatory signalling settled right back down. It's another brick in the wall of "stop the pore, calm the fire," this time in T-cell immunity.
